Western Blot

Western blot หรือเรียกอีกชื่อหนึ่งว่า Immunoblot เป็นเทคนิคที่ใช้ติดตามโปรตีนที่สนใจในสารตัวอย่าง เช่น homogenate tissue หรือโปรตีนที่สกัดมา เป็นวีธีการคล้ายการทำ southern blot hybridization แต่เปลี่ยนจากการหาดีเอ็นเอที่มีความจำเพาะในตัวอย่างมาเป็นโปรตีนแทน ขบวนการในการทำ western blot ประกอบด้วย 6 ขั้นตอนหลักๆ (figure1) ดังนี้

Southern Blotting and Hybridization เป็นเทคนิคทางพันธุวิศวกรรมที่มีความสำคัญและใช้กันอย่างแพร่หลาย ผู้พัฒนาเทคนิคนี้คือ Edward M. Southern ในปี 1975 โดยเขาได้ใช้ดีเอ็นเอที่ติดฉลากด้วยสารรังสีเป็นตัวตรวจจับกับดีเอ็นที่มีความจำเพาะกับ probe ในดีเอ็นเอตัวอย่าง (hybridization) ดังนั้นจึงเรียกเทคนิคนี้ตามชื่อว่า southern blot hybridization
