Overview of ELISA and Detection Strategies

     ในปัจจุบันชุดการทดสอบ Enzyme-linked immunosorbent assay หรือ ELISA นั้นมีให้เลือกใช้ถึง 3 รูปแบบด้วยกัน ซึ่งการเลือกใช้นั้นขึ้นอยู่กับรูปแบบของผลการทดลองที่นักวิจัยต้องการทราบ ซึ่งทั้ง 3 รูปแบบมีรายละเอียดดังนี้

1. Direct Assay เป็นรูปแบบของชุด ELISA ที่ต้องการตรวจหา antigen โดยใช้ primary antibody ที่ติดฉลากด้วยเอนไซม์ เช่น HRP โดยมีลักษณะที่จำเพาะต่อ antigen นั้น หลังจากนั้นเติมตัว substrate เพื่อตรวจสอบสีที่เกิดขึ้น ซึ่งวิธีนี้ไม่ได้รับความนิยมมากนัก
2. Indirect Assay เป็นการใช้ antibody สองตัวในการตรวจสอบ ประกอบด้วย primary antibody และ secondary antibody โดยตัวของ primary antibody จะเข้าไปจับกับ antigen หรือ target ก่อน แล้วหลังจากนั้นใช้ secondary antibody ที่ติดฉลากเอนไซม์เข้าไปจับอีกครั้งหนึ่ง
3. Sandwich Elisa เป็นชุดทดสอบที่ใช้กันอย่างแพร่หลาย หรือ ได้รับความนิยมมากที่สุด เนื่องจากมี sensitivity และ specificity ที่ค่อนข้างสูง เนื่องจากตัวเพลทจะมีการ coated ด้วย antibody ที่จำเพราะต่อ antigen หรือ target ที่สนใจ หลังจากนั้นจะทำการใช้ primary antibody อีกตัวหนึ่งเข้ามาจับกับ antigen นั้น และทำการตรวจสอบด้วย secondary antibody ที่ติดฉลากด้วยเอนไซม์ ในบางครั้งอยากจะติดฉลากด้วย avidin หรือ biotin ที่เป็นตัวขยายสัญญาณ ทำให้สะดวกมากขึ้นในการตรวจตัวอย่างที่มีปริมาณของ antigen ที่ค่อนข้างน้อย

In the assay, the antigen of interest is immobilized by direct adsorption to the assay plate or by first attaching a capture antibody to the plate surface. Detection of the antigen can then be performed using an enzyme-conjugated primary antibody (direct detection) or a matched set of unlabeled primary and conjugated secondary antibodies (indirect detection).
