DNAExtractor Fat is designed for the extraction of high-quality and pure DNA from pure fat and oily food and feed samples as well as from emulsifiers, e.g. lecithin samples. Due to the nature of those sample matrices, there
is no need of a Proteinase K digestion as commonly used, however fat and oily substances are separated from the DNA in a first step before subsequent extraction. The extracted DNA can be used for all kinds of subsequent analytical methods, e.g. real-time and conventional PCR testing.
The DNA isolation process of the DNAExtractor Fat kit is based on an organic extraction using Chloroform extraction and DNA precipitation. Hexane is used to extract fatty and oily substances and subsequently remove those from the aqueous DNA containing phase. Subsequent extraction using Chloroform will further separate potential cell depris from the aqueous DNA containing phase. In the subsequent step DNA is precipitated with isopropanol. The following washing step with ethanol removes remaining reagents and salts that could inhibit the subsequent applications. Afterwards, the DNA is solved in low salt buffer or water to be ready-to-use for subsequent analysis or amplification. Since the composition and biochemistry of the different sample types is very heterogeneous, the extraction of DNA can be a challenging procedure. Depending on the sample type and the subsequent application, further cleaning of the DNA can be required, e.g. with DNA Cleaning Columns (cat. no. 5224700310).
