DNA CLEANING COLUMNS, 100 TESTS
The DNA Cleaning Column kit is designed for the purification of DNA for a broad range of subsequent applications. The kit contains pre-packed and equilibrated ready to use columns, which are very convincing for practical implementation. The procedure is simple and fast and enables the simultaneous procession of numerous samples. DNA Cleaning Columns are optimised for sample volumes of 10 – 100 µL archiving good product yields of DNA with high quality and purity. Since non-specific binding to the matrix components is negligible, samples in the nanogram to milligram range can be purified. Furthermore, the sample is not diluted during the procedure, so no significant reduction of the DNA concentration of the sample occurs. Due to these characteristics DNA Cleaning Columns are ideal for post purification of DNA isolates and have been validated in conjunction with the use of the automated iMAGo setup, e.g. for the removal of potential PCR inhibiting substances prior to real-time PCR analyses.
The DNA purification process of the DNA Cleaning Column kit is based on the principle of size exclusion chromatography in a spin-column format. DNA isolates are load on the top of a matrix consisting of Sephacryl® resin beads, which exhibits a defined pore size. Small molecules can enter these pores, whereas large molecules (e.g. DNA) remain in the space between the matrix beads. During centrifugation large molecules, moving quickly through the matrix were eluted. Smaller molecules were held back in the pores and thus removed from the sample. This technique leads to a fast and efficient purification of DNA molecules from salts, impurities and potential PCR inhibiting substances, without a significant reduction of the DNA content of the sample.
